| Research Interests Combinatorial Biocatalysis The discovery of unique marine isoprenoids has led our group to plan to derivatize these molecules and to assay them as potential anticholesterolemic agents. Thus, I am developping a convenient screen to detect inhibitors of the key enzyme of the cholesterol metabolism, HMG-CoA reductase. This assay done on mouse cells, and based on a LC-MS detection aims at screening agents that would be competitive inhbitors of the enzyme, or that would repress its synthesis through a feedback regulation. I also plan to derivatize these unique marine isoprenoids by enzymatic reactions in both organic solvents and aqueous solution and to use various ources of enzymes either in vivo or in cell-free extracts, in order to obtain a myriad of structures and substitutes. This research interest lies in the new molecules to be synthesized, in new methods of chemoenzymatic derivatization, which have not been used reviously to produce libraries of novel compounds starting from isoprenoids and on the convenience of the assay developed. |
